Today, we will be learning how to take WAINUA — which is used to treat adults with polyneuropathy associated with hereditary transthyretin-mediated amyloidosis, or “hATTR-PN”.

WAINUA is used to treat polyneuropathy (damage to the body’s peripheral nerves) in adults who have stage 1 or stage 2 hereditary transthyretin-mediated amyloidosis (hATTR-PN).

WAINUA comes in a device called an autoinjector. This is what it looks like.

Each WAINUA autoinjector has 1 dose in it. This part at the end is the clear cap. It is on top of the orange needle guard, which covers and hides the needle.

This is the viewing window; you can see the medicine through it.

Before injecting WAINUA, your healthcare professional should show you or your caregiver how to use it.

Step 1 — To get started, gather all the supplies you will need. Have your autoinjector ready. You may need to get your own alcohol wipes, cotton balls or gauze, bandages, and a puncture-resistant disposal container for sharps.

Step 2 — Inspect the carton and wait. If you have been storing your autoinjector in the refrigerator, make sure to let it sit for at least 30 minutes at room temperature before using it.

Step 3 — When you are ready, take the autoinjector out of the carton and inspect it.

Check the expiration date and look for any damage. Make sure the clear cap is attached. Do not use if the expiration date has passed, the autoinjector appears damaged, or the clear cap is missing or not attached.

Take a look at the medicine through the viewing window too.

Make sure the medicine is the right colour. It should be clear and colourless to slightly yellow. If the medicine is cloudy, the wrong colour, or if you see large particles floating in it, do not use it. Call your healthcare professional.

It is normal to see small air bubbles in the liquid.

Step 4 — If everything looks correct, you can now inject WAINUA. You must choose a spot on the lower part of your stomach or the front of your thigh.

If someone is helping you, they can also choose the back of your upper arm. Do not try to inject yourself on the back of the upper arm.

If you will be giving yourself WAINUA, you must choose a spot on the lower part of your stomach (abdomen) or the front of your thigh.

There are a few things to remember when choosing an injection site.

For each injection, choose a different site that is at least one inch (or 3 centimeters) away from where you last injected.

If you choose your stomach, where you inject should be at least 2 inches (or 5 centimeters) away from your belly button.

It is important to choose a spot that is not bruised, tender, red, warm, scaly, or hard.

Do not inject through clothing or into any scars or damaged, discoloured, or tattooed skin.

Today, I will show you an injection on a thigh.

There are a few things to remember when choosing an injection site:

Step 6 — Take off the clear cap by holding the autoinjector in one hand, like this, and pulling the cap straight off with the other hand. Do not twist the clear cap.

Now, the orange needle guard will be exposed. Do not touch or push it or try to recap it. This could cause the medicine to come out before you are ready or damage the autoinjector.

Step 7 — Next, inject using the autoinjector. Place the orange needle guard flat at a 90-degree angle against your skin with the viewing window facing you, so you can see when all the medicine has been injected.

For demonstration purposes, the viewing window is facing the audience instead of the patient.

To inject, firmly press and hold the autoinjector against your skin for 10 seconds. You will hear a click right away. This tells you the injection has started.

Keep the autoinjector still – do not move or reposition it after the injection has started.

You will see the orange plunger start to fill the viewing window.

You may hear a second click during the injection. This is normal. But the injection is not complete until the orange plunger rod completely fills the viewing window.

Step 8 – check the viewing window. Make sure all the medicine has been injected.

If the orange plunger rod does not fill the viewing window, you may not have received the full dose. If this happens or if you have any other concerns, contact your healthcare professional.

Step 9 — Check the injection site. There may be a little blood or liquid. That is normal. If needed, gently press on the area with a clean cotton ball and cover it up with a bandage.

And finally, step 10 — Now that you have injected WAINUA, do not recap the autoinjector or reuse it. Throw the autoinjector into a sharps container right away. It can’t go in the household trash.

Remember, the recommended dose of WAINUA is one 45 mg injection once a month – or the contents of the autoinjector.

The WAINUA autoinjector should be kept in its carton in the refrigerator, but you can store it at room temperature in its carton – up to 30 degrees Celsius – for up to 6 weeks.

You can carry your autoinjector with you; just remember to store it properly. If it is stored outside of the refrigerator for 6 weeks or more, the autoinjector must be thrown away.

Store the autoinjector in the original package to protect it from light. Do not freeze it and do not expose it to heat.
